At least 2 g of biomass is required for representative determination. A larger amount guarantees a more representative result.
The biomass sample is ground with a grinder (homogenization), the appropriate amount is weighed and the sample is then extracted several times with alcohol. The extract or droplets are weighed and diluted with a suitable solvent. The method for determining the content of cannabinoids and terpenes is gas chromatography (GC), in which a flame ionizing detector (FID) is used for detection. The contents are determined by the external standard method, and reference identification solutions are used to identify individual cannabinoids and terpenes.
Quantification limit (LOQ): 200 ppm or 0,02%.
Detection limit (LOD): 100 ppm or 0,01%.
Determination of cannabinoid content using gas chromatography is one of the officially recognized methods. Using gas chromatography with FID detector, we provide a wide linear range of the method and the possibility of detecting compounds without chromophores (i.e. compounds that do not absorb light in the UV-visible range). The contents of related substances (unknown cannabinoids) can also be evaluated, as the response of the detector depends only on the effective number of carbon atoms in the molecule (in HPLC-UV technique the response depends upon the wavelength at which the analysis is performed). The biggest disadvantage of gas chromatography is the inability to separate carboxylated from decarboxylated cannabinoids (without derivatization), because at high temperature during the analysis the decarboxylation process takes place and therefore we can determine only the total content of individual cannabinoids (sum of decarboxylated and carboxylated).
